Do squats make your butt bigger?
Squats can add more lean muscle to your body, so if you're looking to see gains in your butt area, then yes, squats can help with that.
What's the fastest way to get a bigger butt
It will usually take anywhere from two to three months to see results. The first four weeks or so is all pre-conditioning. So it's your muscles adapting to exercise. Probably after those four weeks or so, then the body will transition into developing more lean muscle. It takes two to three months to actually see that, but you will actually feel the muscle tightening.

Work out two to three days a week to develop more muscle, or work out once a week to maintain where you're at.
